Topos Bio – Redefining Druggable Space
Our mission is to unlock a vast, untapped class of disease-relevant targets: Intrinsically Disordered Proteins (IDPs). These proteins, which lack stable structure under physiological conditions, comprise at least 30% of the human proteome and play key roles in a wide array of diseases, including neurodegenerative disorders, cancer, and metabolic dysfunction. Despite their relevance, IDPs have remained largely intractable to traditional drug discovery due to their structural plasticity and the inadequacy of conventional structure-based design tools. Recent advances in AI, such as AlphaFold, have made enormous progress in modeling structured proteins, but these tools fall short when applied to IDPs, which often exist as dynamic ensembles rather than adopting a single defined conformation.
